1. Fortitude Rating = the number printed in the Damage Box of every successfully played card you place in your Ring area. so the ans is yes, your fortitude for the waist lock will be gone. If you want, you could play the throwback version of the waist lock. The TB version does not have the "when this card is in your Ring Area,on your opponents turn, he may discard an applicable Reversal card to put this card into your Ringside pile".
2. Yes you can play them both in the same turn unless the first maneuver didn't got reversed.
